This Application Note presents a comparative study of lung adenocarcinoma (LUAD) and squamous cell carcinoma (LUSC), both subtypes of non-small cell lung cancer (NSCLC), utilizing the Chromium Single Cell Gene Expression Flex assay. The study involved profiling approximately 900,000 cells from 32 human lung donors, focusing on the differences in molecular characteristics and cell-type distributions between the tumor subtypes and normal adjacent tissues. The methodology included sample preparation, hybridization, and sequencing, with a pseudobulk differential gene expression analysis revealing tumor-specific markers associated with poor prognosis. The findings indicate significant variations in cell-type proportions and expression levels of specific markers across the biological groups. The results underscore the utility of the Chromium platform in scaling comparative studies of lung cancer, providing insights into the underlying molecular mechanisms that differentiate LUAD and LUSC, which may inform future therapeutic strategies.
