This technical report presents the advantages of utilizing an automated solid phase extraction (SPE) system integrated with the Agilent 6495D triple quadrupole LC/MS system for the bioanalysis of oligonucleotides. The study specifically focuses on the quantification of mipomersen in pig plasma, demonstrating the high sensitivity and low detection limits achievable with the 6495D system. The automated SPE workstation significantly reduces manual intervention time, enhancing operational consistency and reliability. The report details the complete workflow from sample preparation to data analysis, emphasizing the efficiency of the automated process in comparison to traditional manual methods. Additionally, it discusses the optimization of liquid chromatography conditions and the use of specific reagents to improve detection sensitivity.
