This technical report presents a proof-of-concept study focused on protein biomarker discovery using the Agilent 6495D Triple Quadrupole LC/MS system. The study outlines a rapid and economical workflow that enables the development of comprehensive multiple reaction monitoring (MRM)-based assays without the need for extensive proteome identifications. Utilizing human plasma samples, the report details the methodology employed, including the use of publicly available peptide MS/MS spectral libraries and Skyline software. Key features of the Agilent 6495D system are highlighted, such as improved analytical sensitivity and faster MS acquisition times.
