This case study details the procedures involved in gene therapy and stem cell transplantation via MRI-guided intraparenchymal delivery into brain regions, specifically targeting cortical and subcortical areas. The study outlines two distinct procedures conducted on adult rhesus and cynomolgus macaques, emphasizing the use of real-time MRI imaging to monitor the infusion of treatments. The first procedure involved the placement of an MRI-compatible cannula for the infusion of a paramagnetic contrast agent, allowing for the visualization of the infusate within the targeted brain regions. Serial MRI scans were utilized to ensure proper distribution of the infusate. The second procedure focused on deeper brain regions, with one subject receiving an AAV5 vector mixed with gadolinium for tracking. Post-infusion analysis included hematoxylin and eosin staining and immunostaining for green fluorescent protein, confirming successful delivery into the hippocampus.
