This technical report presents a comparability study of the CFX Touch and CFX Opus Real-Time PCR platforms, focusing on their analytical equivalence. The study is essential for the pharmaceutical and biotechnology industries, as it provides evidence that changes in protocols or equipment do not adversely affect previously approved processes. The report outlines the experimental design, which includes comparisons of specificity, linearity, accuracy, precision, limit of detection, and system suitability across both systems. The studies utilized Zika, Dengue, and Chikungunya assays to test the performance of the CFX Opus 96 and CFX Opus 384 systems against their predecessors. Detailed acceptance criteria were established for both 96-well and 384-well formats, ensuring rigorous testing standards. The results demonstrate intrasystem equivalence, supported by statistical analyses such as Bland-Altman plots and Deming regression. This report serves as a guide for laboratories transitioning to the CFX Opus platforms, ensuring compliance with industry standards.
