This technical report presents a case study on the application of high-throughput peptide purification using the PurePep® EasyClean (PEC) technology in drug discovery. The objective is to address the challenge of low-purity crude peptides often used in screenings, which can lead to unreliable assay results. The study details the synthesis of 96 peptides in parallel using solid-phase peptide synthesis (SPPS) and the subsequent purification process adapted to a 96-well plate format. The report outlines the methods employed for peptide synthesis, purification, and analysis, including the use of a vacuum manifold for efficient processing. Results indicate a significant increase in mean purity from 68% to 87% after purification, demonstrating the effectiveness of PEC technology in removing impurities. The findings highlight the potential for improving assay reliability by minimizing peptide-related impurities, thus facilitating the development of peptide drugs.
