This technical report outlines the ENRICH-iST workflow designed to enhance the analysis of plasma proteomes by enriching low abundant proteins. The report describes how the technology effectively reduces the dynamic range of protein abundance in biofluids, allowing for the identification of over 2000 proteins in human plasma samples, with more than 1000 proteins quantified reliably. The methodology includes sample preparation using the iST kit and the ENRICH-iST kit, followed by analysis on the TimsTOF HT mass spectrometer. The study involved plasma samples from non-small cell lung cancer patients and healthy donors, demonstrating the ability to distinguish between different patient groups based on protein profiles. The report presents data showing that the ENRICH-iST method retains sample-specific protein abundance ratios while significantly increasing the number of detectable proteins compared to neat plasma samples.
