This technical report presents a protocol for the extraction and purification of genomic DNA (gDNA) from whole blood using Covaris' AFA-TUBE TPX Plate and Adaptive Focused Acoustics (AFA) technology. The study evaluates the efficiency of DNA extraction from samples of 12 healthy donors, demonstrating recoveries ranging from 376 to 809 ng with an average fragment size exceeding 1 kb. The protocol is designed to streamline high-throughput processes while reducing hands-on time and eliminating the need for specialized equipment like vacuum manifolds. The entire workflow can be completed in less than 90 minutes, and the extracted DNA is shown to be free of PCR inhibitors, with purity levels assessed using A260/A280 ratios. The report also discusses the repeatability of the extraction process, with coefficient of variance values for DNA yields within replicates being less than 10%. Additionally, the potential for high molecular weight DNA extraction is noted, which can be critical for various downstream applications, including next-generation sequencing.
