This technical note presents a detailed comparison between enzymatic DNA synthesis (EDS) and chemical DNA synthesis (CDS) processes, focusing on the capabilities of the SYNTAX platform developed by DNA Script. The document outlines the inherent challenges associated with traditional phosphoramidite-based CDS, including accessibility, environmental impact, turnaround time, and efficiency. It highlights how the EDS process overcomes these limitations by enabling consistent, on-demand production of high-quality DNA oligos in a standard laboratory environment. The study evaluates oligos of two lengths, 60-mers and 120-mers, comparing delivery times, purity, and sequence accuracy between CDS and EDS. Results indicate that EDS significantly outperforms CDS in terms of delivery time and consistency, while maintaining comparable sequence accuracy. The findings underscore the advantages of EDS in producing oligos efficiently and sustainably, marking a shift in the standards for DNA synthesis in molecular and synthetic biology applications.
