This application note presents the integration of a ProCellics Raman Analyzer into a bioprocess control system for glucose monitoring in CHO cell cultures. The study outlines the implementation of a feedback control loop utilizing OPC UA connectivity to maintain glucose concentration at a steady level of 5 g/L over a period of three days. This automation minimizes the need for human intervention, reducing labor costs and contamination risks associated with manual sampling. The document details the experimental setup, including the bioreactor control system and feeding strategies, as well as the methodologies used for monitoring glucose levels. Results indicate that the Raman Analyzer provided accurate glucose measurements, leading to improved process performance and lower lactate production compared to traditional fed-batch cultures. The findings support the use of Raman spectroscopy for in-situ monitoring and control in biopharmaceutical manufacturing, aligning with Process Analytical Technologies (PAT) and Quality by Design (QbD) principles.
