This application note presents a technical report on the comparative analysis of media types and volumes for optimizing plasmid yield using the AmMag™ Quatro Mini 1100 automated purification system. The report outlines the challenges associated with traditional manual plasmid purification methods, which are labor-intensive and prone to variability. It details the automated workflow of the AmMag™ Quatro Mini 1100, which employs alkaline lysis and magnetic bead-based purification to streamline the process. The study assesses plasmid yield and purity across different E. coli growth conditions, specifically focusing on various media types including LB, TB, and a proprietary Plasmid+ medium. The results indicate that TB medium consistently produced the highest plasmid yields, while the purity was evaluated through absorbance measurements. Additionally, the report compares the performance of the automated system to a manual miniprep kit, highlighting the efficiency and quality of plasmid purification achieved with the AmMag™ Quatro Mini 1100.
