This research article presents a meta-analysis of randomized controlled clinical trials focused on understanding the drivers of placebo response in atopic dermatitis (AD). The study aims to quantify factors influencing the placebo effect, particularly examining the role of protocol design, including the use of topical anti-inflammatories. The analysis includes 68 studies that featured a placebo arm where the Eczema Area and Severity Index (EASI) 75 was measured. The findings indicate that placebo rates are notably higher in trials that permit the use of topical anti-inflammatory agents, with a significant increase in placebo rates observed over time. The research employs a multivariate linear regression model to analyze various predictors, such as study start date and patient demographics. The results suggest that the increasing trend in placebo rates may be linked to the allowance of topical treatments and the timing of study initiation, accounting for a substantial portion of the variance in the data. Future research directions are also discussed.
