This technical report presents a study on the efficiency of DNA extraction from archaeological and recent skeletal remains using an automated protocol on the EZ2 Connect Fx instrument. The study addresses the challenges faced by scientists in obtaining reliable DNA information from degraded skeletal remains, which often contain minimal DNA and are susceptible to contamination. The report outlines the methodology employed, including the use of silica-coated magnetic bead technology, and compares it to traditional organic extraction methods. A total of 16 skeletal samples were processed, including nine archaeological teeth samples and seven recent femoral bone fragments. The results indicate a range of DNA concentrations across samples, with the automated method proving effective in extracting DNA while minimizing contamination risks. The study concludes that the EZ1&2 DNA Investigator Kit and EZ2 Connect Fx instrument represent valuable tools for analyzing skeletal remains, particularly for well-preserved samples.
