This application note presents a comparative evaluation of two mixing systems, the 10 L ProConnex MixOne Carboy and the 10 L Tulip tank with a magnetic stirrer, in the context of tangential flow filtration (TFF) processes in biopharmaceutical manufacturing. It outlines the critical role of efficient mixing in ensuring the quality and consistency of products, particularly in achieving uniform distribution of active pharmaceutical ingredients (APIs). The study employs the CTech FlowVPX in-line analytical system to measure protein concentration continuously during ultrafiltration/diafiltration processes. Results indicate that both mixing systems achieved sufficient homogeneity of products, with the ProConnex MixOne Carboy demonstrating advantages for shear-sensitive materials. The analysis includes operational parameters such as power efficiency and shear, emphasizing the importance of real-time concentration measurements for process effectiveness. The findings support the integration of advanced mixing technologies to enhance downstream processing performance in the biopharmaceutical sector.
