This white paper presents the Resilience 3-day short cycle CAR-T manufacturing process, which significantly reduces the time required to generate activated CAR-T cells compared to the conventional 7-day process. The shorter cycle not only minimizes the time, materials, and labor needed for production but also enables a faster release of CAR-T cells for patient treatment. The paper details the high purity, activation, and transduction rates of the T-cells produced by this method, highlighting their younger phenotype and enhanced biological activity. The results indicate that the 3-day process generates CAR-T cells that exhibit improved IFN-γ release and cytotoxic activity against target cells, even at lower effector to target ratios. Additionally, the 3-day process demonstrates a lower effective dose requirement for patients, approximately 2 log-fold lower than the conventional method.
