This technical report discusses the application of advanced tools in Next Generation Risk Assessment (NGRA) for cosmetic ingredients. The report details the use of advanced organ models, including microphysiological systems (MPS), as refinement tools when lower tier approaches are insufficient. It presents the applications of MPS in NGRA, such as individual organ systems and multiorgan-on-a-chip models, to investigate toxicity mechanisms and organ communication. The evaluation of a toolbox for assessing chemical-exposure scenarios is documented, highlighting its protective capabilities against high-risk scenarios and identifying limitations in current models. Additionally, the report addresses the implementation of various testing strategies, including a human liver 2 compartment metabolizing system and body-on-chip technology, to enhance drug risk assessment and repurposing.
